Just replaced all the red and blue (hot/cold) valves with new valves on the manibloc, cause one of the "diamond" shaped knobs broke.
My question is do you put thread sealant on them when re attaching them ( i know tape thread sealant is a no no) and what kind do you use if indeed you use it on the plastic fitting. Any suggestions would be appreciated.

You don't need anything on the threaded end where the PEX fitting connects.
I did not put anything on mine.

White Permatex with Teflon.
The RV tech that did mine used Teflon water pipe sealant (not tape). Been working well for three years. Stuff Roger recommended would also be a proven sealant. My Manablock had a crack so total replacement.
If by "re attaching" you mean the Pex lines going to each fixture, they should incorporate a cone washer that does the sealing.
I would replace the cone washers- I always replace them if fitting has been in place for a while--I have had problems in the past when reusing fittings with old cone washers. They are not easy to find but you can get them on amazon Cirque Bg/10 1/2" Saniprene Seal 06435

New cone washers on mine never any thread sealant. If you have a leak with new washers, look for cracked fitting or leaking line connection
I also replaced my cone washers. The old one were deformed from age.
No sealant, just silicone grease on the rubber cone, and DON"T overtighten!!!
HOWEVER, before you twist off one of your manabloc valve diamond thingys, and you eventually will....order one of these.

They fit the old manablocs perfectly and even comes with a handle and a tool to pop the old valve body out and you just snap in the new one. I also got several 3 pack valve bodies on Amazon, without the tools and will eventually replace all of them. Woody.
before I get started replacing the cone washers, mine are in pretty rough shape, can someone explain which way to install them? small end towards the hose or large end?
Re-plumbed my entire house several years back with pex and manifold. Use no sealant at manifold. Used sharkbites at fixtures. Not one problem.
Large end goes in first said another way when it is in and you look at the hose in your hand you should be looking at the taper end
